Billy Mure naît à New York. Il débute par le violon à l'âge de cinq ans, il poursuit à la guitare à sept ans et à treize ans il commence à composer. Pendant la seconde guerre mondiale, il dirige une formation musicale de l'Air Corps. Ensuite, après la guerre, il joue dans une station de radio jusqu'en 1957.
Il devient par la suite musicien de session, arrangeur, compositeur.
Il est connu pour des albums très particuliers : 'Super-Sonic Guitars In Hi-Fi' en 1957 pour RCA où il utilise le multipiste pour créer des effets à plusieurs guitares. Dans son album suivant pour RCA 'Fireworks', il explore les sons "pyrotechniques" en Living Stereo.
Il enregistre aussi des albums de guitare hawaiienne en y intégrant des percussions inhabituelles.
